Hello

If I turn my key in the door lock and there is no resistance in either direction (and locked door doesn't open), is it most likely the cylinder that needs to be replaced?

Maybe one of the plastic actuator rod clips broke and the rod fell out of place . the tumblers must be ok if you can put the key in and turn it. sleek

Sounds like one of the retainers has fallen off and the rod fell out. You can buy the retainers from Dorman or used from Ebay.

Either #1 or #3 fell off

The rod may have also fell off the latch part as well if the clip broke (or is missing).
