Mithridatism is the practice of protecting oneself against a poison by gradually self-administering non-lethal amounts. The word derives from Mithridates VI, the King of Pontus, who so feared being poisoned that he regularly ingested small doses, aiming to develop immunity.
Memorably encountered as a stratagem used by a maniacal KGB agent (John van Dreelen) to dupe a cornered but determined Russian defector (Martin Landau) into drinking a glass of wine laced with a knockout drug in "The Jeopardy Room", Season 5, Episode 149, The Twilight Zone, hosted by Rod Serling, first airing on April 17, 1964.

According to some historians, the official food taster for Pope Alexander VI (Rogrigo Borgia) did the same thing in order to attempt to administer a deadly dose to the Pope in 1501. The attempt reportedly failed and the taster was himself poisoned in the process. Pope Alexander is believed to have died of malaria in 1503.

On Mithridatism, I've heard of snake handlers doing the same thing with snake venom. They would inject small doses of snake venom and gradually raise the doses till they would be completely immune to the venom to the poisonous snake they would be handling.
